I want to load a sound file on to a blank V5 Direct Board. Are there any steps that I need to take to ensure that the sound file isn’t erased if I have to do a factory reset at some point?

Sound files for ESU LokSound decoders cannot be erased AFAIK. They can be over-written or modified but will not be affected by a decoder reset. The decoder reset only resets CVs to the decoder's default settings. Note that these default settings can be changed when programming CVs with the LokProgrammer. This is an option when writing changes to the decoder.
If the sound file has multiple prime movers, horns or bells it is a good idea to update the decoder defaults when writing your choices to the decoder, otherwise you will have to re-select them after doing a decoder reset.

Early versions of the software didn't have the box for overwrite as new default checked as default. If it was written that way, a reset wouldn't be a reset to what you loaded. Now, that box is automatically ticked by default, so your upload will automatically become the latest reset point.
